Glutathione Isn't a Supplement. It's a Loop.

Somewhere in your medicine cabinet, or one click away in your cart, is a bottle of glutathione capsules. The logic is airtight: glutathione is your master antioxidant, you want more of it, so you swallow some. Thirty dollars, problem solved.
Except the body doesn't work like a shopping cart. Most of that glutathione never arrives where you wanted it, and the reason why is the single most useful thing you can understand about how this molecule actually operates. Once it clicks, the entire supplement aisle starts to look a little different.
What actually happens when you swallow glutathione
Glutathione is a small protein — a tripeptide, three amino acids strung together: cysteine, glycine, and glutamate. Your digestive system is very good at exactly one thing when it meets a small protein: taking it apart. Enzymes in the gut snip the bonds and break most of that intact glutathione back down into its three separate amino acids before it ever crosses into your bloodstream.
So you paid for finished glutathione and, in practical terms, absorbed a scattering of raw parts. Raising the glutathione level inside your cells — which is the only place it does anything — by swallowing the finished molecule is, at best, unreliable. Your cells would rather build it themselves, on-site, from ingredients they can actually import. Which they do, constantly, as long as they have what they need.
That's the first myth gone. Here's the bigger one.
Glutathione lives in two forms, and only one of them works

Picture your cell's glutathione not as a stockpile but as a workforce that clocks in and out. Active glutathione — the form written as GSH — goes out and neutralizes a free radical. In the act of doing so, it gets chemically "spent": two of them bond together into an oxidized, inactive form written as GSSG. Clocked out. Useless in that state.
If that were the end of the story, you'd burn through your entire glutathione supply in minutes. It isn't the end of the story. Your cell has a recovery step — an enzyme called glutathione reductase — that takes the spent GSSG and flips it back into two active GSH. Clocked back in. Ready for the next radical.
The amount of glutathione you have barely matters. What matters is the ratio of clocked-in to clocked-out — how much of it is actually available to work right now.
This is why a lab could measure your "total glutathione" as perfectly normal while you feel like garbage. Total glutathione counts everyone on the payroll, clocked in or out. If most of your pool is stuck in the spent GSSG form because the recovery step has stalled, you have plenty of glutathione and almost none of it working. The stockpile is fine. The workforce is on break.
The recovery step runs on a fuel called NADPH
So everything hinges on that recovery enzyme. What does it need to do its job? Two things, and neither of them is more glutathione.
First, it needs a fuel called NADPH — the molecule that carries the electrons used to flip spent glutathione back to active. No NADPH, no recovery, full stop. Your cells make NADPH through a specific route called the pentose phosphate pathway, and the throughput of that pathway is governed in part by thiamine (vitamin B1). Run low on the pathway's support nutrients and your NADPH supply tightens, which starves the recovery step no matter how much raw glutathione is standing around.
Second, the recovery enzyme itself is built to run on riboflavin (vitamin B2) — specifically its active form, which sits inside glutathione reductase and makes the whole reaction possible. The enzyme without its riboflavin cofactor is a machine without a power cord.
Read that back. The thing standing between you and a working antioxidant system isn't the antioxidant. It's two B vitamins and a metabolic pathway most people have never heard of. This is the part the glutathione-capsule crowd skips entirely.
Why NAC beats a glutathione pill
Now the logic inverts cleanly. If your cells build glutathione on-site, the smart move isn't to import the finished molecule — it's to hand your cells the one ingredient they tend to run short on and let them build all they need.
Of the three amino acids in glutathione, two are usually easy to come by. Glutamate is everywhere. Glycine is common. The bottleneck is almost always cysteine — it's the scarce one, the rate-limiter, the ingredient your production line runs out of first. Supply cysteine reliably and glutathione synthesis speeds up. Leave it short and it doesn't matter what else is on the shelf.
This is exactly what NAC (N-acetylcysteine) does. It's a stable, well-absorbed delivery vehicle for cysteine — it survives the trip, gets into your cells, and gives them the missing building block to make their own glutathione, intracellularly, where it counts. You're not importing the finished product and hoping it survives customs. You're supplying the raw material and letting the factory run.
But — and this is the whole point of the article — substrate alone isn't enough. Handing the factory more cysteine while the recycling loop is stalled is like delivering lumber to a construction site with no power tools. You need both: the material to build fresh glutathione, and the working recovery loop to keep the glutathione you've got in its active form. Miss either half and the system limps.
The crew, seat by seat

This is why a serious glutathione formula looks less like a big scoop of antioxidant and more like a short list of very specific parts, each holding one seat in the same machine:
- NAC — supplies cysteine, the rate-limiting building block, so your cells can synthesize fresh glutathione.
- Glycine (delivered as magnesium glycinate) — the second building block in the glutathione molecule, plus magnesium as a broad metabolic cofactor.
- Vitamin B6 (as P5P) — drives the pathway that generates cysteine from its precursors, backing up your supply of the scarce ingredient.
- Riboflavin (as R5P) — the cofactor seated inside the recovery enzyme itself; without it, spent glutathione can't be flipped back to active.
- Thiamine (as benfotiamine) — supports the pentose phosphate pathway that produces NADPH, the fuel the recovery step burns.
- Molybdenum — clears sulfite, the byproduct left over when your body processes all this sulfur-rich cysteine, so the exhaust doesn't back up.
- Vitamin C — runs alongside glutathione in the antioxidant network, where the two help regenerate and spare each other.
Notice what's not on that list: a giant dose of finished glutathione. You don't need it. You need the material to build your own and the working loop to keep recycling it.
One loop, one system
That's the reasoning behind RE-BALANCE, so we'll keep this short. It isn't a pile of antioxidants and it isn't a glutathione pill. The bottom line
Swallowing glutathione mostly feeds a gut that breaks it apart before it arrives. The molecule works in a loop — active, spent, restored — and the thing that stalls is the recovery step, which runs on NADPH and B vitamins, not on more glutathione. The efficient fix is to supply cysteine (via NAC) so your cells build their own, and supply the cofactor crew that keeps the loop recycling. Material plus loop. Skip either half and it limps.
